{"id":470,"date":"2023-09-01T22:21:28","date_gmt":"2023-09-01T22:21:28","guid":{"rendered":"https:\/\/pokharelsugam.com.np\/?p=470"},"modified":"2023-10-02T16:07:22","modified_gmt":"2023-10-02T16:07:22","slug":"prepare-temporary-slide-of-onion-cell-practical","status":"publish","type":"post","link":"https:\/\/pokharelsugam.com.np\/school-science\/prepare-temporary-slide-of-onion-cell-practical\/","title":{"rendered":"TO PREPARE TEMPORARY SLIDE OF  ONION CELL FROM EPIDERMAL PEEL OF ONION BULB"},"content":{"rendered":"\n<h2 class=\"wp-block-heading\">OBJECTIVES<\/h2>\n\n\n\n<ul class=\"wp-block-list\">\n<li>To prepare temporary slide.<\/li>\n\n\n\n<li>To Show Plant Cell<\/li>\n\n\n\n<li>To Show cells of onion<\/li>\n<\/ul>\n\n\n\n<h2 class=\"wp-block-heading\">MATERIALS REQUIRED<\/h2>\n\n\n\n<p>Fresh onion bulb, Safranin, Dropper, Glycerol, No. 0  painting round tip brush, forceps, needle, blade, scissor, wash  bottle, petri dish, watch glass, cover glass, slide, compound microscope .<\/p>\n\n\n\n<h2 class=\"wp-block-heading\">THEORY<\/h2>\n\n\n\n<p>Onion cells are commonly used for microscopic studies because they have large and easily observable cells. The onion cell has a rectangular shape and is enclosed by a cell wall. Inside the cell, you can find a nucleus and cytoplasm. Preparing a temporary slide of an onion cell allows us to observe these cellular structures under a microscope.<\/p>\n\n\n\n<h2 class=\"wp-block-heading\">PROCEDURE<\/h2>\n\n\n\n<ul class=\"wp-block-list\">\n<li>Obtain a fresh onion bulb, that is free from damage or disease.<\/li>\n\n\n\n<li>Peel a thin, transparent layer from the onion bulb using a scalpel or razor blade. Try to get a layer that is as thin as possible.<\/li>\n\n\n\n<li>Cut the ventral peel in square about 3-4 mm length by scissor then keep in watch glass containing distilled water.<\/li>\n\n\n\n<li>Then transfer peel through brush to another watch glass containing 2-3 drops of Safranin and wait for 30 to 60 seconds.<\/li>\n\n\n\n<li>Then wash stain by transferring tissue to another watch glass containing clean distilled water for 2 or 3 times to remove stain completely.<\/li>\n\n\n\n<li>Take a clean microscope slide and pour a drop of glycerin at the center of slide.<\/li>\n\n\n\n<li>Then transfer tissue to slide containing glycerine.<\/li>\n\n\n\n<li>Then apply cover slip with needle to prevent bubble.<\/li>\n\n\n\n<li>Then watch under the compound microscope with 10X objective at first then at 45X objective.<\/li>\n\n\n\n<li>You find many stomata with guard cell, then draw diagram in practical note.<\/li>\n<\/ul>\n\n\n\n<h2 class=\"wp-block-heading\">OBSERVATION<\/h2>\n\n\n\n<ul class=\"wp-block-list\">\n<li>The onion cells are typically rectangular or square in shape.<\/li>\n\n\n\n<li>A distinct cell wall surrounds each cell.<\/li>\n\n\n\n<li>A large central nucleus is visible within the cell.<\/li>\n\n\n\n<li>The cytoplasm fills the space between the cell wall and the nucleus.<\/li>\n\n\n\n<li>The iodine stain, if used, highlights the nucleus.<\/li>\n<\/ul>\n\n\n\n<h2 class=\"wp-block-heading\">RESULT<\/h2>\n\n\n\n<p>A temporary slide of an onion cell has been successfully prepared and observed under the microscope.
